Precision and scaling in morphogen gradient read-out
Morphogen gradients infer cell fate as a function of cellular position. Experiments in Drosophila embryos have shown that the Bicoid (Bcd) gradient is precise and exhibits some degree of scaling. We present experimental results on the precision of Bcd target genes for embryos with a single, double or quadruple dose of bicoid demonstrating that precision is highest at mid-embryo and position dependent, rather than gene dependent. This confirms that the major contribution to precision is achieved already at the Bcd gradient formation. Modeling this dynamic process, we investigate precision for inter-embryo fluctuations in different parameters affecting gradient formation. Within our modeling framework, the observed precision can only be achieved by a transient Bcd profile. Studying different extensions of our modeling framework reveals that scaling is generally position dependent and decreases toward the posterior pole. Our measurements confirm this trend, indicating almost perfect scaling except for anterior most expression domains, which overcompensate fluctuations in embryo length

Exogenous schwann cells migrate, remyelinate and promote clinical recovery in experimental auto-immune encephalomyelitis
Schwann cell (SC) transplantation is currently being discussed as a strategy that may promote functional recovery in patients with multiple sclerosis (MS) and other inflammatory demyelinating diseases of the central nervous system (CNS). However this assumes they will not only survive but also remyelinate demyelinated axons in the chronically inflamed CNS. To address this question we investigated the fate of transplanted SCs in myelin oligodendrocyte glycoprotein (MOG)-induced experimental autoimmune encephalomyelitis (EAE) in the Dark Agouti rat; an animal model that reproduces the complex inflammatory demyelinating immunopathology of MS. We now report that SCs expressing green fluorescent protein (GFP-SCs) allografted after disease onset not only survive but also migrate to remyelinate lesions in the inflamed CNS. GFP-SCs were detected more frequently in the parenchyma after direct injection into the spinal cord, than via intra-thecal delivery into the cerebrospinal fluid. In both cases the transplanted cells intermingled with astrocytes in demyelinated lesions, aligned with axons and by twenty one days post transplantation had formed Pzero protein immunoreactive internodes. Strikingly, GFP-SCs transplantation was associated with marked decrease in clinical disease severity in terms of mortality; all GFP-SCs transplanted animals survived whilst 80% of controls died within 40 days of disease
Uncovering the topology of configuration space networks
The configuration space network (CSN) of a dynamical system is an effective
approach to represent the ensemble of configurations sampled during a
simulation and their dynamic connectivity. To elucidate the connection between
the CSN topology and the underlying free-energy landscape governing the system
dynamics and thermodynamics, an analytical soluti on is provided to explain the
heavy tail of the degree distribution, neighbor co nnectivity and clustering
coefficient. This derivation allows to understand the universal CSN network
topology observed in systems ranging from a simple quadratic well to the native
state of the beta3s peptide and a 2D lattice heteropolymer. Moreover CSN are
shown to fall in the general class of complex networks describe d by the

RELIABILITY OF A DIGITAL METHOD TO DETERMINE FRONTAL AREA OF A CYCLIST
Eight cyclists were photographed with a digital camera for three trials while positioned on their own bicycle wearing their helmet. The positions were different from each other and
described as: hands on the brake hoods; hands below the curve of the brakehoods on the handlebars; and using aerobars. Twenty four trials were digitized by two different
individuals three times to estimate the inter- and intra-rater reliability of the method. The Intraclass Correlation Coefficient (p < 0.05) value for the intra-rater (test-retest) reliability was ICC = .993; for inter-rater consistency the ICC = .976. There were significant differences (p < 0.05) between digitizers and between trials apparently due to a learning
effect that disappeared by the third trial. Due to small differences between digitizers and trials, caution is recommended when considering use of this method
On the criticality of inferred models
Advanced inference techniques allow one to reconstruct the pattern of
interaction from high dimensional data sets. We focus here on the statistical
properties of inferred models and argue that inference procedures are likely to
yield models which are close to a phase transition. On one side, we show that
the reparameterization invariant metrics in the space of probability
distributions of these models (the Fisher Information) is directly related to
the model's susceptibility. As a result, distinguishable models tend to
accumulate close to critical points, where the susceptibility diverges in
infinite systems. On the other, this region is the one where the estimate of
inferred parameters is most stable. In order to illustrate these points, we
discuss inference of interacting point processes with application to financial
data and show that sensible choices of observation time-scales naturally yield

Dietary salt intake assessed by 24 h urinary sodium excretion in Australian schoolchildren aged 5–13 years
Objective To measure total daily salt intake using 24 h urinary Na excretion within a sample of Victorian schoolchildren aged 5–13 years and to assess discretionary salt use habits of children and parents.Design Cross-sectional study.Setting Completed within a convenience sample of independent primary schools (n 9) located in Victoria, Australia.Subjects Two hundred and sixty children completed a 24 h urine collection over a school (34 %) or non-school day (66 %). Samples deemed incomplete (n 18), an over-collection (n 1) or that were incorrectly processed at the laboratory (n 3) were excluded.Results The sample comprised 120 boys and 118 girls with a mean age of 9·8 (sd 1·7) years. The average 24 h urinary Na excretion (n 238) was 103 (sd 43) mmol/24 h (salt equivalent 6·0 (sd 2·5) g/d). Daily Na excretion did not differ by sex; boys 105 (sd 46) mmol/24 h (salt equivalent 6·1 (sd 2·7) g/d) and girls 100 (sd 41) mmol/24 h (salt equivalent 5·9 (sd 2·4) g/d; P = 0·38). Sixty-nine per cent of children (n 164) exceeded the recommended daily Upper Limit for Na. Reported discretionary salt use was common: two-thirds of parents reported adding salt during cooking and almost half of children reported adding salt at the table.Conclusions The majority of children had salt intakes exceeding the recommended daily Upper Limit. Strategies to lower salt intake in children are urgently required, and should include product reformulation of lower-sodium food products combined with interventions targeting discretionary salt use within the home

Enteric infectious diseases in swine have considerable economic impact on the industry due either to mortality, cost of treatment or reduced growth rates which can lead to extended production periods. The consequences can be catastrophic especially in nursery sites since young piglets are in a susceptible period associated with immature immune system and are often affected by rapid dehydration related to neonatal diseases
